HD is a mid-life onset neurodegenerative caused by an abnormal polyglutamine repeat in the N-terminal of the huntingtin (htt). We have recently shown that htt is phosphorylated by Cdk5 at serines 1181 and 1201 (S1181 and S1201). These phosphorylations can be induced by DNA damage in vitro and in vivo. Absence of phosphorylation of htt at S1181 and S1201 confers toxic properties to wild-type htt in a p53-dependent manner in striatal neurons and accelerates neuronal death induced by DNA damage. In contrast, phosphorylation at serines 1181 and 1201 protects against polyQ-induced toxicity. We propose to further analyse the underlying mechanisms in cellular models. In addition, the objective of this thesis will be to characterize the physiological roles of these phosphorylations.
